Based on a conditionally regulated and modular gene vector system, pCON, we pursue two parameters in detail: (i) Directed targeting of extrachromosomal vectors to predetermined regions of the nucleus (or even a single gene) will address the question, how functional chromatin (or even a single genetic locus) affects the genetic stability of a gene vector and its capacity for gene expression.
