Epigenetic of Vector-Chromatin Interactions
After transduction of extrachromosomal gene vectors, initial establishment and genetic stability of vector DNA are the critical parameters for reliable gene expression. Based on a conditionally regulated and modular gene vector system, pCON, we pursue two parameters in detail: (i) Directed targeting of extrachromosomal vectors to predetermined regions of the nucleus (or even a single gene) will address the question, how functional chromatin (or even a single genetic locus) affects the genetic stability of a gene vector and its capacity for gene expression. (ii) We will analyze the efficiency with which extrachromosomal gene vectors become established after transduction of DNA and focus on the superior efficiency of virally transduced gene vectors.
